【Animal modeling】-Phagocytosis experiment of mouse peritoneal macrophages

Purpose requirements 1. Understand the principle of phagocytosis of mouse peritoneal macrophages 2. Familiar with the basic process of phagocytosis Fundamental The phagocytosis of

cells is a way of ingesting nutrients in unicellular animals. Macrophages, monocytes and neutrophils in higher animals have phagocytic function and are an important part of the body's non-specific immune function. Macrophages are generated by differentiation of bone marrow stem cells. When pathogenic microorganisms or other foreign bodies invade the body, macrophages will accumulate at the foreign body due to their chemotaxis. The macrophages can internalize them into the cytoplasm to form phagocytic vesicles. , And then fuse with lysosomes intracellularly to digest and decompose foreign matter. The formation of phagocytic vesicles requires the help of microfilaments and their binding proteins. If the cells are treated with the drug cytochalasin B that degrades microfilaments, the formation of phagocytic vesicles can be blocked; the immunosuppressant cyclophosphamide (CYP) can affect macrophages. Phagocytosis of cells also has an impact.

Experimental supplies 1. Equipment: microscope, dissecting dish, scissors, forceps, syringe, glass slide, cover glass, pipette, etc. 2. Reagents:

0.85% normal saline; Alsever blood preservation solution; 6.0g of soluble starch and 100ml of water, boil for later use.

  3. Material: mice; 1% chicken red blood cell suspension

  Methods and steps

After blood is taken from the lower vein of the chicken wings, add normal saline and centrifuge twice, then add appropriate amount of normal saline according to the volume of red blood cells obtained to adjust the concentration of chicken red blood cells to 1%.

Three days before the experiment, 1ml of soluble starch was injected into the abdominal cavity of the mice every day, and 1ml of 1% chicken red blood cells were injected into the abdominal cavity 30 minutes before the experiment.
