【 Animal Modeling 】 - Evaluation of Syndrome in a Tidal Heat Model Rat Induced by Bilateral Ovariectomy and Tamoxifen Administration

  Objective: To explore a syndrome animal model for studying women's hot flashes.

  Method: 24 female SD rats were randomly divided into a sham surgery group, a bilateral ovariectomy group, and a tamoxifen group, with 8 rats in each group. Bilateral oophorectomy and gavage of 10 mg/(kg · d) tamoxifen were used to simulate two types of hot flashes animal models, respectively. On the 14th and 28th day of modeling, characterization information such as open field activity, anal temperature, and surface infrared thermography of rats were detected. On the 29th day, the rats were euthanized, and the uterus was weighed and pathological sections were made; ELISA detection of blood levels of estradiol and catecholamines; RT qPCR was used to detect the gene expression of adrenal sex hormone synthase (Star, Cyp11a1, Cyp17a1, Cyp19a1, Por, Hsd3b2, Hsd17b1) and adrenal medullary catecholamine synthase (Th, Ddc, Dbh, Pnmt).

  Compared with the sham surgery group, the weight gain of rats in the bilateral ovariectomy group was significant (P<0 01), while the rats in the tamoxifen group showed slow weight gain. The highest surface temperature of rats in the bilateral ovariectomy group significantly decreased on the 28th day (P<0 01), similar to the manifestation of Yang deficiency, and the difference between the highest and lowest abdominal temperatures significantly increased on the 14th day (P<0 The difference between the highest and lowest back temperatures significantly increased on the 28th day (P<0. 05) 01), similar to the internal heat phenomenon, and with a decrease in open field activity in rats (P<0 01), similar to the manifestation of insufficient vital energy; After 14 days of administration, the highest surface temperature of rats in the tamoxifen group significantly decreased (P<0 01), but the open field activity of rats increased (P<0 01). Both modeling methods resulted in a significant decrease in rat uterine index (P<0 01). Compared with the sham surgery group, both the bilateral ovariectomy group and the tamoxifen group showed a significant decrease in E2 (P<0 01), NE and Epi both showed a decreasing trend (P<0 05), bilateral ovariectomy group β- EP also significantly decreased (P<0 05); Compared with the sham surgery group, the expression of Cyp11a1 gene in the adrenal gland of bilateral ovariectomized rats significantly increased (P<0 05), while the expression of Cyp17a1 and Hsd17b1 genes significantly decreased (P<0 05); The expression of Star and Por genes in the tamoxifen group significantly increased (P<0 01), while the expression level of Cyp17a1 gene significantly decreased (P<0 01); The expression of Pnmt gene was significantly downregulated in the bilateral ovariectomy group (P<0 01).

  Conclusion: Bilateral ovariectomy in SD rats can be used for the study of perimenopausal hot flashes, and its syndrome characteristics are similar to traditional Chinese medicine's kidney yang deficiency combined with yin deficiency and internal heat.
