[Animal Modeling] - Establishment of Calcitonin Gene Related Peptide Transgenic Mice and Dynamic Analysis of Blood Pressure

Purpose: To establish calcitonin gene related peptides α and β Transgenic mice were used to dynamically compare and analyze their blood pressure, and an animal model suitable for hypertension research was established.

Method: Convert human CGRP α And CGRP β Gene insertion into chickens separately β- Downstream of actin promoter, construction of transgenic expression vector, establishment of C57BL/6J CGRP by microinjection method α And CGRP β Transgenic mice, PCR identification of transgenic mouse genotypes. Identification of CGRP using Western Blot α And CGRP β Screening for high expression transgenic strains in heart, lung, kidney, and liver tissues. Analyze the dynamic blood pressure changes in transgenic mice using a non-invasive blood pressure measuring instrument.

Result: High expression of CGRP was established in the heart, lungs, kidneys, and liver tissues α CGRP of genes α Transgenic mice; High expression of CGRP in lung and liver tissues β CGRP of genes β Transgenic mice. CGRP α Transgenic mice with normal blood pressure and CGRP β Transgenic mice exhibit significant blood pressure reduction at 12 months of age.

Conclusion: CGRP α And CGRP β Transgenic mice can be used as animal models for studying the mechanisms of peripheral vascular resistance and hypertension resistance in CGRP.
