Objective: To establish an atherosclerotic model of phlegm and blood stasis in rats, and observe the changes of inflammatory factors in the model rats and the intervention effect of the prescription (Danlou Tablet).
Method: 32 male SD rats were randomly divided into normal group, model group, atorvastatin calcium group (ATV group), and Danlou group (DLP group), with 8 rats in each group. The normal group rats were fed with basic feed, while the remaining three groups were all treated with a high-fat diet+intraperitoneal injection of vitamin D3 combined with carotid artery balloon injury to replicate the phlegm stasis type AS model. The model group, ATV group, and DLP group were respectively given physiological saline, atorvastatin calcium suspension, and Danlou tablet suspension by gavage intervention for 4 weeks. After the intervention, HE staining was used to observe the pathological changes in the thoracic aorta of rats, and to detect blood lipids, hs CRP, IL-6, and TNF in the serum of rats- α Detection of IL-6 and TNF in rat thoracic aortic tissue using RT-PCR with LP-PLA2 levels- α And LP-PLA2 mRNA expression.
Result: ① Compared with the normal group, the serum levels of TC, LDL-C, hs CRP, IL-6, and TNF in the model group rats were higher- α Elevated levels of LP-PLA2 (P<0.05), decreased levels of HDL-C (P<0.05), and IL-6 and TNF in vascular tissues- α The expression of LP-PLA2 and LP-PLA2 increased (P<0.05). Under the HE staining light microscope, the endothelial cells of the thoracic aorta in the model group were incompletely arranged, the intima was significantly thickened irregularly, the space under the endothelium was widened, foam cells were gathered, and lipid was deposited, forming a typical AS plaque. ② Compared with the model group, the serum TC, LDL-C, hs CRP, IL-6, and TNF of rats in the ATV group and DLP group were compared- α Reduced levels of LP-PLA2 and IL-6, TNF in thoracic aortic tissue (P<0.05)- α The expression of LP-PLA2 decreased (P<0.05), and="" there="" was="" no="" statistically="" significant="" difference="" between="" the="" atv="" group="" dlp="" p="">0.05). Under HE staining light microscope, the intimal hyperplasia, plaque area and degree of atherosclerosis of thoracic aorta in ATV group and DLP group were significantly reduced.
Conclusion: The method of using a high-fat diet+intraperitoneal injection of vitamin D3 combined with carotid balloon injury can establish a rat model of AS with phlegm stasis syndrome, and the phlegm stasis treatment formula (Danlou tablets) can improve the AS and inflammatory response of the model rats.
