Objective: To explore the influencing factors of establishing a C57BL/6J mouse melanoma lung metastasis model, including tumor inoculation method, cell inoculation quantity, and tumor formation cycle
Method: Cultivate mouse melanoma cells B16F10 in vitro; 1) Eighteen male mice aged 6-8 weeks were randomly divided into three groups, with six mice in each group. The mice were injected intravenously, intraperitoneally, and subcutaneously, with each group receiving 100 injections μ L (3 × 106 cells) B16F10 cell suspension, after 2 weeks, dissect mice and observe the growth and metastasis of melanoma; 2) Divide into three groups, as above, and inject 3 × 106 cells, 1 × 106 cells, and 3 × 105 cells respectively through the tail vein. After 2 weeks, dissect the mice and observe the growth and metastasis of melanoma; 3) Divide into three groups, as above, inject 1 × 106 cells into the tail vein, dissect the mice at 1 week, 2 weeks, and 3 weeks, and observe the growth and metastasis of melanoma
Result: 1) After tail vein injection of mouse melanoma cells, the success rate of lung metastasis in mice was 100%, while intraperitoneal and subcutaneous injection did not cause lung metastasis; 2) When the number of melanoma cells inoculated into mice was 1 × 106, the number of melanoma cells that experienced lung metastasis was moderate; When the number of inoculated cells is 3 × 106, there is an excessive number of melanoma cells that undergo lung metastasis; When the number of inoculated cells is 3 × 105, the number of melanoma cells that undergo lung metastasis is relatively small; 3) After injecting 1 × 106 mouse melanoma cells into the tail vein and feeding for 2 weeks, significant lung metastasis of melanoma cells can be observed without causing mouse death; After 3 weeks of feeding, the number of melanoma cells metastasizing to the lungs was excessive, and more than half of the mice died; After one week of feeding, the number of melanoma cells metastasizing to the lungs is relatively small
Conclusion: The recommended method for constructing a C57BL/6J mouse melanoma lung metastasis model is to inject 1 × 106 mouse melanoma cells into the tail vein and grow for 2 weeks
