[Animal Modeling Pharmacodynamic Evaluation] - Rat Pulmonary Aspergillus Infection Model

  (1) Method of modeling: Adult SD rats were instilled with Aspergillus fumigatus suspension through tracheal intubation to establish a rat lung infection model. The suspension of Aspergillus fumigatus was inoculated into the rats through nasal and oropharyngeal instillation methods to establish an oropharyngeal colonization model.

  (2) Result: Pathological examination of lung tissue: Aspergillus mycelium was found in 80% of infected rat lung tissue, and there were no significant abnormalities under the light microscope in the lung tissue of the colonization group and healthy group rats; Plasma β - glucan detection results: The plasma β - glucan concentrations in rats infected with Aspergillus fumigatus were (34.1 ± 12.8) pg/ml, respectively. The plasma β - glucan concentrations in healthy and colonized groups were (12.2 ± 5.5) pg/ml and (12.8 ± 6.7) pg/ml, respectively. The comparison of Aspergillus fumigatus infection with healthy and colonized groups was P<0.01. The G test method can be used to detect the concentration of plasma β - glucan as a diagnostic indicator for pulmonary fungal infection, with a diagnostic threshold of 20pg/ml, which is more sensitive and specific.
