[Animal Modeling Pharmacodynamic] Evaluation Kidney Deficiency Syndrome Animal Model

  The kidney is the foundation of congenital diseases. Kidney deficiency syndrome is one of the main types of syndromes in traditional Chinese medicine, mainly including kidney yang deficiency, kidney yin deficiency, kidney qi deficiency, and kidney essence deficiency. And kidney yang is the primary yang of human beings. The vital activities of various organs in the human body depend on the warmth and promotion of kidney yang. The animal model of kidney yang deficiency is currently the earliest and most widely studied syndrome model, with many modeling methods.

  (1) Housing overwork law

  According to the theory of traditional Chinese medicine that excessive labor in the room leads to kidney deficiency, excessive fatigue and frequent mating in mice can create an animal model of kidney deficiency.

  【 Modeling Method 】 Female mice were subcutaneously injected with long-acting contraceptive injections. After about 3 days, they entered the estrus phase, which can be confirmed by vaginal smear examination of keratinocytes. Then, the male mice were co housed with an estrus female mouse with a vaginal plug for 1 day, and then co housed with 5 estrus female mice. It is important to replace the estrus female mice in a timely manner. Male mice swim once a day in the morning and afternoon, and are pulled out when they are unable to continue swimming and are about to sink.

  【 Model characteristics 】 It was observed that the animals gradually developed symptoms of kidney yang deficiency such as fear of cold, lethargy, and lack of movement. After 4 weeks, they were euthanized and organs such as the thymus, adrenal gland, and testes were quickly removed for relevant indicator measurements. Animal weight, testicular weight, thymus weight, total activity of testicular lactate dehydrogenase, and relative activity of lactate dehydrogenase isoenzyme X all decreased.

  Model evaluation and application: The modeling factors of this model are more in line with the causes of traditional Chinese medicine kidney deficiency syndrome. The kidney deficiency syndrome model has been widely used in pathological research on kidney deficiency syndrome and aging, as well as pharmacological research on kidney tonifying traditional Chinese medicine.

  (2) Estradiol benzoate method

  【 Modeling Mechanism 】 This drug induces inhibition of the hypothalamic pituitary gonadal axis in mice, resulting in a kidney yang deficiency model. Animals exhibit signs of reduced activity and decreased fatigue resistance.

  【 Modeling Method 】 ICR mice were intraperitoneally injected with 0.4mg/mm benzoic acid estradiol soybean oil diluted solution at a dose of 0.1ml/10g, once a day. Inject continuously for 15 days to create a kidney yang deficiency model. Measure the physical signs, anal temperature, grip, swimming time, autonomous activity, serum creatinine and urea, hematological indicators, testicular and seminal vesicle index of mice.

  【 Model features 】 The model mice have wet and sweaty fur standing upright, mental lethargy, arched back atrophy, and prefer to squeeze and lie down rather than move. The anal temperature significantly decreased, swimming time decreased, mouse grip weakened, number of autonomous activities decreased, serum creatinine and urea increased, red blood cell parameters increased, white blood cell parameters decreased or remained unchanged, and testicular and seminal vesicle index decreased.

  【 Model Evaluation and Application 】 This model introduces modern medical detection methods and indicators to make research more objective and reliable, and is widely used in pharmacological research on kidney yang deficiency.

  (3) Thyroidectomy method

  【 Modeling Mechanism 】 Metabolism in individuals with hypothyroidism generally slows down, and the basal metabolic rate decreases. Most patients with hypothyroidism based on traditional Chinese medicine have symptoms of spleen kidney yang deficiency. Therefore, the animal model of "yang deficiency" caused by removing animal thyroid can simulate human yang deficiency syndrome.

  【 Modeling Method 】 Splague Dawley rats weighing between (280 ± 20) g were raised at room temperature, with controlled air humidity and free access to water and food. Remove the thyroid gland from the rat and continue feeding.

  【 Model characteristics 】 After 7 days, rats showed symptoms of kidney yang deficiency such as eating and drinking less, reducing body weight, slow response, reduced activity, arched back hair, dull hair color, easy shedding, and loose stools.

  【 Model Evaluation and Application 】 This model can be combined with differential proteomics techniques to study the liver mitochondrial proteome of kidney yang deficiency rats and investigate the pathogenesis of kidney yang deficiency rats.
