1. Modeling material animals: dogs, weighing over 10kg; Medications: pentobarbital, physiological saline, lidocaine; Equipment: Animal artificial ventilator.
2. Modeling method: After anesthesia with pentobarbital, the animal is fixed in the supine position, and the trachea is cut open and connected to the animal's ventilator for artificial respiration. The chest is opened between the left fourth rib to expose the heart, and the pericardium is cut. A silk thread is threaded at the upper 1/3 (1-2cm from the starting point of the anterior descending branch), 1/2, or 1/3 (above the apical branch) of the left coronary artery to ligate it, causing myocardial infarction. This is the first stage ligation method. To reduce the occurrence of arrhythmia and death in animals, the two-stage ligation method is often used, which involves threading two silk threads at one ligation point, placing the No. 5 needle parallel to the coronary artery in the knot, ligating the first silk thread (first knot), ligating the needle together with the anterior descending branch of the coronary artery, and withdrawing the needle; After 30 minutes, ligate the second silk thread (second ligation); At this point, the left anterior descending coronary artery is completely ligated. If epicardial electrodes are not used, the pericardium can be sutured, the chest wall can be sutured to restore natural breathing, and an electrocardiograph can be connected to observe changes in the electrocardiogram. Physiological saline can be dripped from the femoral vein and external jugular vein according to changes in blood loss.
3. The modeling principle is to replicate the myocardial infarction model by ligating the coronary artery.
4. Pathological changes after modeling: The electrocardiogram of the animal showed ST segment elevation and pathological Q waves, indicating the occurrence of myocardial infarction.
5. Precautions: Surgical instruments should be strictly disinfected to prevent surgical infections, surgical trauma should be minimized, and aseptic operation should be strictly enforced.
