【Animal Modeling 】Drug Efficacy Evaluation Deoxycorticosterone Acetate Hypertension Model

  1. Modeling material animal: rat, female, 100-150g; Drug: Deoxycorticosterone acetate or valerate, NaCl.

  2. Model making method: Remove the left kidney of the model animal, and then drink 1% NaCl water. Inject deoxycorticosterone acetate or valproate subcutaneously into rats at a dose of 20-30 mg/(kg · w), which can be injected several times a week or once a week. The methods of adding surfactants to physiological saline and embedding DOCA tablets subcutaneously in animals can also be used.

  3. The modeling principle involves the use of high doses of DOCA (a precursor of aldosterone) during the modeling process, which has similar physiological effects to aldosterone. It feedback inhibits the circulating renin-angiotensin system, leading to decreased plasma renin activity. Therefore, it is a low renin type hypertension model.

  After about 2 weeks of modeling, the blood pressure of the animals begins to rise, and malignant hypertensive disorders appear after 5-7 weeks.
