[Animal Modeling - Pharmacological Evaluation ]- Animal Model of Kidney and Kidney Yang Deficiency Syndrome Caused by Labor Injury

  (1) The replication method takes male mice as the modeling objects and uses healthy estrus period female mice to induce "atrioventricular agenesis". After subcutaneous injection of 0.05ml of long-acting contraceptive injection into female mice, the mice entered the estrus period after about 3 days. The degree of keratinization was confirmed by vaginal smear examination. Each male mouse was co housed with one female mouse. The next day, male mice with vaginal plugs were taken out of the cage and kept on a regular diet. Each male mouse is housed in the same cage as 5 estrus females, and attention should be paid to timely replacement of estrus females. For 4 consecutive weeks, swim once in the morning and once in the afternoon each day, and remove each time until they are unable to continue swimming and about to sink. Record the time. Observe the general condition of the animals, and after 4 weeks, euthanize the animals. Weigh the thymus, adrenal gland, and testes, and measure the total activity of lactate dehydrogenase and lactate dehydrogenase isoenzymes in testicular homogenate.

  (2) Model characteristics: As time goes on, animals gradually show symptoms of kidney deficiency such as lethargy, fear of cold, less arching, slow response, crowding together, reduced diet, dull fur, obvious vertical hair, damp abdominal fur, scrotal folds, and testicular recovery. Animals exhibit obvious kidney deficiency, and their body weight, testicular weight, thymus weight, total activity of testicular lactate dehydrogenase (LDH), and relative activity of LDH-X all decrease significantly, with a tendency to reduce adrenal gland weight.

  (3) Comparative medicine, based on the theory of excessive fatigue and lack of joints in traditional Chinese medicine, utilizes the colligel effect of rodents to induce frequent mating. At the same time, it combines forced swimming, injection of methylprednisolone, or fear of kidney injury to create a model of kidney (yang) deficiency syndrome. The molding process introduces two factors: fatigue (or fear) and labor, and there are few reports of using labor factors alone for molding. The process of damage to male mice with kidney yang deficiency caused by atrioventricular disorders is a chronic process, and mice have strong reproductive ability, so it takes a long time to cause kidney yang deficiency. This method is less commonly used.
