1. Modeling material animal: monkey, weighing 2.2-2.4kg; Drug: glomerular basement membrane, complete Freund's adjuvant.
2. Modeling method
(1) Preparation of GBM: GBM from normal humans, rabbits, rats, and dogs is placed in isotonic saline and stirred in a stirrer for 3-5 minutes to make a homogenate containing 38-50mg of glomeruli per ml. 1:10000 thiomersal is added and mixed with 85ml of complete Freund's adjuvant to form a 1:1 water oil emulsion containing approximately 19-25mg/dl GBM.
(2) Modeling: Select healthy monkeys and feed them in metabolic cages. Inject 2ml of GBM once in the second week, divided into two parts, and inject 1ml intramuscularly into each part. If injected subcutaneously or intradermally, divide the total amount of 2ml into 5 injections. Regularly check urine protein, blood urea nitrogen, serum albumin, etc. before injection and during immunization. Inject 3 to 24 times.
3. Modeling principle: Induction of animal glomerulonephritis model by heterologous immune complexes.
4. Changes after modeling: After modeling, most animals showed proteinuria 21-36 days after injection, and blood urea nitrogen significantly increased within 19-61 days. Histological examination showed that the cortical structure of the monkey kidney was damaged, manifested by epithelial cell proliferation, occasional crescent shaped glomeruli, significant glass like changes, and white blood cell infiltration in some glomeruli. Renal tubules dilate, with polymorphonuclear leukocytes, red blood cells, and crystals, accompanied by fibrosis and monocyte infiltration in the interstitium.
