3. The principle of modeling is to establish a chronic renal failure and hypertension animal model by feeding rats with adenine for a long time.

As the number of days of adenine solution gavage increases, the water intake of the model group increases, with the highest water intake reaching 2-3 times that of the normal control group; At the same time, weight loss, significant increase in urine output, dry and dull body hair, mental lethargy, and in severe cases, muscle tremors, convulsions, exhaustion, and death may occur.
After 7 weeks, the model group showed a significant decrease in red blood cells (RBC), hemoglobin (Hb), total protein (TP), and albumin (ALB), while systolic blood pressure (SBP), total cholesterol (TC), serum urea nitrogen (Urea), creatinine (Crea), and urine protein/creatinine continued to increase, with significant differences compared to the normal control group.
The appearance of the kidneys in the control group rats was not swollen, with a glossy red color, and the capsule was easy to peel off. The morphology of the glomeruli and renal tubules was normal, and the boundary between the cortex and medulla was clear. The kidneys of the model group rats were significantly enlarged, with both kidneys appearing gray white and granular on the surface. The adhesion between the renal capsule and renal parenchyma was difficult to peel off, and the boundary between the cut surface and the medulla was unclear. Light microscopy examination: The number of glomeruli decreased, with an average of 2-6 per low magnification field (normal 6-8), showing atrophy and a basically normal morphology. Cystic dilation of renal tubules, extensive crystalline deposition in the renal cortex, and foreign body giant cell reactions can be seen around the crystals. A large number of renal tubules are destroyed, atrophied, and disappear. There is infiltration of inflammatory cells. Interstitial fibrosis is evident.
