[Animal Modeling - Pharmacological Evaluation] - Partial Simple Epilepsy Model in Rats

  1. Modeling material animals: rats, male or female, weighing 200-300g; drugs: pentobarbital, FeCl3 solution; Equipment: Rat brain stereotaxic device.

  2. Modeling method: After intraperitoneal injection of 1% pentobarbital (30mg/kg) to anesthetize rats, fix them on a rat brain stereotaxic device. After routine disinfection, cut open the scalp and drill a 2-3mm diameter cranial foramen at 2mm behind the coronal suture and 3mm beside the midline. Radial incision of the dura mater is made; Fix FeCl3 solution containing 100mmol/L onto a directional micro thruster using a 0.5mm diameter PE tube, with the distal end of the tube in contact with the cerebral cortex; The positive pole of the 100 μ A DC electrophoresis apparatus is connected to FeCl3 solution in the PE tube, and the negative pole is connected to the ipsilateral temporal muscle through the acupuncture and moxibustion needle, and the electrophoresis is conducted with electricity for 10 minutes; Drill 0.5mm bone holes 1mm in front of the coronal suture, 0.5mm beside the midline on one side, and 1mm in front of the herringbone suture, and 3mm on both sides of the midline. These three holes are respectively screwed with flat head screws with a diameter of 0.5mm and a length of 5mm to contact the surface of the dura mater, serving as EEG recording electrodes; Immediately after electrophoresis, record the electroencephalogram (EEG) for 3 hours, which is the acute phase EEG; The EEG recorded 15 and 30 days after electrophoresis is the chronic phase EEG.

  3. The principle of modeling is that iron has an epileptic effect. This model is equivalent to acute or chronic partial simple seizures, similar to the onset and pathological changes of traumatic epilepsy.

  4. Changes after modeling: The acute phase EEG epileptic discharge rate is about 75%. After 0.5 hours of electrophoresis, epileptic waves appear on the epileptic side, gradually intensifying. After 1 hour of electrophoresis, the epileptic waves on the opposite side reach their peak at about 1.5 hours and tend to stop around 2 hours. During the chronic phase, all animals' EEG showed epileptic like discharges.

  5. Precautions: Surgical instruments should be strictly disinfected to prevent surgical infections, surgical trauma should be minimized, and aseptic operation should be strictly followed; When drilling a skull, attention should be paid to the depth. Once there is a sense of breakthrough, stop immediately to prevent excessive bleeding.

After surgery, rats should be separated and housed in cages 3-5 hours after awakening to prevent the first waking rats from licking and biting the wounds of comatose rats. Try to maintain the optimal temperature and humidity for rats in the breeding room as much as possible.
