The molding method involves marking the improved polyethylene conduit with built-in threading; Rats were anesthetized by intraperitoneal injection of 6% chloral hydrate (0.4ml/100g), fixed in a dorsal position, and disinfected with 2% iodine and 75% ethanol in the middle of the neck. A midline incision was made in the neck, and the left common carotid artery was separated and threaded for future use; Simultaneously separate the right common carotid artery and thread it for backup; Separate the left internal and external carotid arteries, transect the external carotid artery, ligate the pterygoid artery, cut the opening near the bifurcation of the external carotid artery, insert the catheter into the internal carotid artery, and slowly advance to an appropriate depth; According to the marked length, withdraw the sheath appropriately to allow blood to flow into the catheter for self coagulation, and slowly push in 20U of thrombin. Causing blood clotting and thrombus formation; Insert the trocar again and push it forward evenly and gently to the marked position, so that the formed thrombus blocks the MCA entry into the skull; Indwelling catheter, suturing skin, single cage feeding. The sham surgery group did not insert thrombi and formed blood clots. During the experiment, the anal temperature of the rats was maintained at (37.0 ± 0.5) ℃ and the room temperature was maintained at (26 ± 1) ℃.
3. The modeling principle used rat autologous thrombus combined with suture occlusion to successfully prepare a cerebral ischemia model.
4. Changes after modeling: The brain tissue moisture content of the model group rats was significantly higher than that of the sham surgery group. The cerebral infarction area in the model group rats was significantly increased compared to the sham operation group.
5. Pathological changes after modeling: In the sham surgery group, the neuronal cell membrane was clear and intact, without deformation or edema, and the nucleolus was clearly visible; The brain tissue lesions in the suture group and thrombus combined suture group of rats showed loose interstitium, significant swelling and deformation of nerve cells, unclear cell membranes, deep staining of cytoplasm, nuclear condensation, autolysis and necrosis of nerve cells, significant interstitial edema, and increased gaps around cells and blood vessels.
6. Precautions: Surgical instruments should be strictly disinfected to prevent surgical infections, surgical trauma should be minimized, and aseptic operation should be strictly enforced.
